Lion's Mane Benefits for Mood and Anxiety

Evidence: Emerging — two small human trials, both positive, neither large

Two of the most cited human studies in the entire category are about mood rather than memory. They are small, they are positive, and they are not a treatment for anything.

What was measured

In a 2019 study, seventy-seven people with overweight or obesity took three 400 mg capsules daily for eight weeks and reported reductions in depression, anxiety and sleep-disorder scores. Separately, a four-week trial in menopausal women using lion's mane cookies found lowered depression and anxiety scores.

Cookies, in a clinical trial, sounds like a joke. It is not — it is a food-matrix delivery study, and it is one of the better-controlled pieces of work in the field.

The proposed mechanism

Two threads. The first is anti-inflammatory: chronic low-grade inflammation is increasingly implicated in depression, and lion's mane has demonstrated anti-inflammatory activity. The second is hippocampal — NGF supports neurons in the hippocampus, a region involved in regulating emotional response, and reduced hippocampal neurogenesis is one of the better-supported biological correlates of depression.

Both trials were small, short, and in specific populations. Consistent direction across two independent studies is worth noting. It is not the same as an established treatment.

What this does not mean

  • It is not an antidepressant and must not replace one. Stopping prescribed medication is dangerous.
  • Neither trial studied clinical depression as a diagnosis.
  • If you are on an SSRI or any psychiatric medication, talk to your prescriber before adding anything.
  • Persistent low mood is a reason to see a doctor, not to buy a supplement.
